Yellow represents residues involved in communication between the main active site and regulatory sites (box 22, intermediate entropy and intermediate variability). The green area holds residues involved in regulation (box 23, high entropy and intermediate variability). Blue represents residues with no known function (box 33, high entropy and high variability). In an actual EV-plot, one will find crosses inside each box that represents the residues in the consensus sequence of the MSA
